This is a work in progress. Here's what I've got so far: (Note: In my campaign, we've decided that the Time War never happened, so Gallifrey and the Time Lords still exist.)
In the First Doctor sourcebook, one of the adventure hooks involves the tooth that the Doctor left behind in Tombstone in 1881 in "The Gunfighters," and it's tie-in to what River said in "The Impossible Astronaut."
"A Time Lord's body is a miracle. Even a dead one. There are whole empires out there who'd rip this world apart just for one cell."
So, one of the Rani's experiments (Still sorting out details.) resulted in her catching a virus that causes rapid aging and isn't purged by regeneration. Her next body just continues to grow old and die within a month. She's already been through three bodies since we last saw her as a result, but now she's found a cure. Unfortunately, it requires the genetic material of a Time Lord under 500 years old who hasn't regenerated yet. Since she can't return to Gallifrey, she'd normally be doomed if she hadn't been able to use an old contact to access Matrix records and discover the Doctor's tooth in Tombstone. Unfortunately, she wasn't the only person her source sold the information to, and two alien races (Still working on which ones they're going to be or if I'll invent new ones.) have found out about the tooth and are also converging on Tombstone in early 1882. (Or December 1881 if you want a Christmas special adventure.)
A complication is that the Rani's aging virus is spreading and the citizens of Tombstone are starting to rapidly age and die. It seems that no race is immune, as the two alien species have also contracted the virus as well. In addition to having to work with The Rani to find a cure, the PCs will also have to determine which, if either, race is entitled to the cure as well.
