Radium blue is a radoactive isotope not found on earth. is used by timelords to help with post regenartion problems. it is highly adictive will kill a human in seconds

A post regeneration "problem" sounds like an interesting plot point to run a session over (especially if nobody can agree on who gets to play the Time Lord - an incapacitated NPC makes a good option here).
Players could start the session having narrowly escaped a violent conflict, with their Time Lord having just regenerated with some difficulty. The TARDIS manages to navigate to a location where Radium Blue is available (albeit, not easily so) with a message from the Time Lord explaining that they need to obtain the substance to restore the Time Lord to consciousness and continue on their journey.
Perhaps the problem could be a form of illness that manifests itself during regeneration - like a sort of cancer, but worse. The Time Lord takes some measures to keep the matter from getting worse (a stasis field, a time loop, self-induced anesthesia, etc) and having foreseen the issue, already programmed a TARDIS protocol complete with holographic message to explain to the Companions what needs to be done.
Of course, Radium Blue being a rare and powerful substance, it doesn't come cheaply. Some points of conflict the characters might have to deal with include: -The substance is being used by a warlord to construct a devastating weapon. Security is tight, and innocent lives will be lost if the weapon isn't taken down. -The substance is the prized possession of a wealthy merchant who requires equally valuable payment in exchange for the Radium Blue - naturally, the payment is something he already has in mind. It might include indentured servitude of one of the Companions (with the deal that whoever sells themselves to the merchant gets to take control of the Time Lord in the next session), a morally questionable transaction (you can have it, if you kill my competitor), or a difficult thing to obtain (the toothbrush of Rasillon is said to be in this cave, guarded by creatures and traps long forgotten even by the Time Lords! Get it for me, and the Radium Blue shall be yours.) -The Radium Blue could be in the possession of a Dalek vehicle or base. The characters will have to take it from the Daleks without being exterminated. The Daleks have probably already detected the TARDIS and will attempt to use the characters to gain access and exterminate the Time Lord)

Well, a Sphere of Annihilation could be done up with a very Whovian feel to it.
